Fluorescence Lifetime Imaging -FLIM

For doing fluorescence lifetime imaging microscopy -FLIM we offer the complete system LIFA -Lambert Instruments Fluorescence lifetime imaging Attachment allowing fluorescence image acquisition and the generation of lifetime images. Additionally the key components of this system, the modulated image intensifier II18MD and the modulated LED light source, are offered as separate products for users who decide to build their own FLIM -fluorescence lifetime imaging microscopy system including their own software.

The LIFA -Lambert Instruments Fluorescence Attachment for lifetime imaging microscopy can be attached to any widefield fluorescence microscope and works in the frequency domain, which means that it uses a modulated light source and a modulated image intensifier as detector. The system is easy to install (within 1 hour) and to operate and does not require an optical bench with laser. The LIFA -Lambert Instruments Fluorescence Attachment system has been judged "easy and highly quantitative". And last but not least, it's reasonable priced.

Confocal or TIRF -Total Internal Reflection Fluorescence LIFA
Typically, the LIFA -Lambert Instruments Fluorescence Attachment is a widefield FLIM -fluorescence lifetime imaging microscopy system with a modulated LED light source. However, other modulated light sources like diode lasers can also be used to perform FLIM -fluorescence lifetime imaging microscopy. This makes the LIFA -Lambert Instruments Fluorescence Attachment suitable not only for widefield lifetime images, but also for confocal lifetime images. Confocal microscopy is an imaging technique used to increase micrograph contrast and/or to reconstruct three-dimensional images by using a spatial pinhole to eliminate out-of-focus light in specimens that are thicker than the focal plane.

II18MD, modulated image intensifier

The II18MD is an Image Intensifier with an 18mm diameter input screen that can be Modulated in the range of 10-120 MHz and is Digital controlled. The II18MD modulated image intensifier is used as key component for FLIM -Fluorescence Lifetime Imaging Microscopy in the frequency domain. The II18MD modulated image intensifier is designed for users who wish to apply the highest quality detector as part of their set-up for fluorescence lifetime imaging.

Spectrally resolved LIFA
Spectrally resolved LIFA
Typically, the LIFA -Lambert Instruments Fluorescence Attachment is a widefield FLIM -fluorescence lifetime imaging microscopy system with a modulated LED light source. The detector is a modulated ICCD and generates 2D images. However, by placing a spectrograph in the light path just prior to the detector the emission light of one line of the image is spectrally resolved providing extra information about the fluorescence. An image is generated with the fluorescence intensity profile at the y-axis and the emission wavelength at the x-axis. The pseudo colored map indicates the fluorescence lifetime.

Modulated LED
Modulated LED

The Modulated LED light source can be used as the excitation light source in FLIM –fluorescence lifetime imaging microscopy systems that operate in the frequency domain. Modulated LEDs of different wavelength bands are available. The output light intensity can be varied over a wide range and the maximum excitation power available at the sample is comparable with that of other light sources.

The ISS Chronos time-resolved fluorometer is a heterodyne frequency-domain instrument based on laser diodes or light emitting diodes (LEDs) and PMT detectors. It has a full automation of instrument components including cuvette holder, polarizers, shutters, filterwheel, monochromators and stirrers. The lifetime measurement capabilities range from milliseconds to picoseconds.

Alba system for confocal imaging

The ISS Alba imaging workstation is a computer-controlled instrument specifically designed for confocal imaging applications, by using scanning mirrors or a piezo-controlled xyz stage. The modular design of the instrument allows it to be easily upgraded to acquire Fluorescence Lifetime data (FLIM) and Fluorescence Correlation Spectroscopy (FCS) data.
