FCS
In confocal Fluorescence Correlation Spectroscopy, the temporal fluctuations of the detected fluorescence signals are recorded. These are the time-dependent spontaneous intensity fluctuations of the fluorescence signal in the observation volume. As the intensity fluctuations in the volume under observation may arise from diffusive Brownian motion, random flow patterns, and chemical reactions, the data stream contains information about these processes occurring at the molecular scale. The fluctuations can be recorded as FCS or as Photon Counting Histogram (PCH), the former technique providing information on the characteristic diffusion times of the molecule and their number, the triplet state function, the size of the excitation volume; while the latter technique recording the brightness of the molecular species and their number.
